dc.description.abstractThis study aimed to develop a methodology of pacu (Piaractus mesopotamicus) embryos cryopreservation and to analyze the injury cells caused in these embryos after the cryoprotectants intracellulares, methanol and ethylene glycol applications, in concentrations of 7%, 10% and 13% with the addition of an extracellular cryoprotector, the sucrose 0,1 M. After the exposure 10 embryos were processed in historesin and the morphologic preservation was evaluated in agreement with three categories considering the characteristics: chorion, yolk, yolk syncytial layer and germ layers. The used cryoprotectors did not alter the chorion preservation. The sucrose had significant effect on yolk preservation when compared to the intracellulares cryoprotectors. A higher acidophilia of the yolk granules indicated a positive effect only when the sucrose was used and with the interaction of cryoprotector intracellular and sucrose. There was positive effect with the use of the sucrose for the number and size characteristics of nuclei present in the yolk syncytial layer with significant effect of the interaction between intracellular cryoprotector and sucrose on the nuclei size. The intracellular cryoprotector ethylene glycol allowed a higher differentiation of the germ layers when compared to the methanol, but there was not any significant difference in using sucrose. The association of the ethylene glycol in the concentration of 7% with sucrose showed to be the best cryoprotectors combination to the preservation of the yolk and germ layers of Piaractus mesopotamicus. To analyzed embryonic morphology a total of 20 embryos/treatment were fill and frozen and after thawing three embryos, on average, resultants of that process were included in historesin, stained with hematoxilin-eosin and analyzed under optical microscope (OM). A straw/treatment containing embryos was thawed and maintained in incubator with the objective in order to accompany the development, not resulting in embryos outbreak. Samples of two embryos submitted to the treatments with methanol and ethylene glycol in the concentration of 10% associated or not with sucrose and the treatments water and water with sucrose frozen and not frozen were processed and analyzed through scanning electron microscopic (SEM). In spite of the embryos presenting the typical structures of the phase such as chorion, yolk, yolk syncytial layer and germ layers they were altered in 100% when the embryos were analyzed under OM and SEM. The chorion became irregular and broken; absence of individualization of the yolk granules; yolk syncytial layer presented shape thickness and irregular size and the blastoderm presented altered nuclei in the shape and sometimes lacking, located in atypical areas or absent in some embryos. No treatment was effective for embryo preservation and consequent appearance. The used protocols did not avoid the intracellular ice formation resulting in serious morphologic alterations during the freezing and thawing processes, making unfeasible the embryos of P. mesopotamicus.en

dc.description.resumoobjetivo nesse trabalho foi desenvolver uma metodologia de criopreservação de embriões de pacu (Piaractus mesopotamicus) e analisar as injúrias causadas nestes embriões após a aplicação de crioprotetores intracelulares, metanol e etilenoglicol nas concentrações de 7%, 10% e l3% e a adição de um crioprotetor extracelular, a sacarose 0,1 M, durante a exposição de 10 minutos, submetidos as técnicas de congelação e descongelação. Após a exposição aos crioprotetores foram processados l0 embriões em historesina e avaliou-se a preservação morfológica de acordo com trêscategorias para as estruturas embrionarias e extra-embrionárias. os crioprotetores utilizados não alteraram a preservação do córion. A sacarose teve efeito positivo sobre a preservação do vitelo quando comparada aos crioprotetores intracelulares. Maior acidofilia dos grânulos de vitelo indica efeito positivo apenas com a utilização de sacarose e interação do crioprotetor intracelular e sacarose.Houve efeito positivo com a utilização da sacarose para as características de número e tamanho dos núcleos presentes na camadas incicial vitelina, com efeito significativo da interação crioprotetor intracelulare sacaÍose sobreo tamanho dos núcleos' o crioprotetor intracelular etilenoglicol permitiu maior diferenciação das camadas germinativas comparado ao metanol, mas não houve diferença significativa de utilização da sacarose.A associação do etilenoglicol na concentração de 7% com a sacarose mostrou ser a melhor combinação de crioprotetores para preservação do vitelo e camadas germinativas de P. mesopotamicus.para a análise da morfologia embrionária foram envasados e congelados 20 embriões/tratamento e após a descongelação três embriões, em média, resultantes desse processo foram incluídos em historesina, corados com hematoxilina-eosina e analisados em microscópio óptico (Mo). um palhete/tratamento contendo embriões foi descongelado e mantidos em incubadora com o objetivo de acompanhar o desenvolvimento , não resultando em eclosão de embriões. Amostras de dois embriões submetidos aos tratamentos com metanol e etilenoglicol na concentração de l0% associado ou não a sacarose e os tratamentos água e água com sacarose foram processados e analisados em microscopia eletrônica de varredura (MEV). Apesar dos embriões apresentarem as estruturas típicas da fase como,córion, vitelo, camadas inicial vitelina e blastoderme, as mesmas encontravam-se alteradas em 100%dos embriões quando analisadas em Mo e MEV. O córion apresentou-se irregular e rompido; ausência de individualização dos grânulos de vitelo; a camadas incicial vitelina apresentou formato, espessura e tamanho irregulares e a blastoderme apresentou núcleos alterados no formato e às vezes ausentes, localizada em regiões atípicas ou ausentes em alguns embriões. Nenhum tratamento foi efetivo para preservação do embrião e conseqüente eclosão. Os protocolos utilizados não evitaram a formação de gelo intracelular resultando em alterações morfológicas graves durante os processos de congelação e descongelação, inviabilizando os embriões P. mesopotamicus.pt_BR
