Identificação das linhagens genéticas principais de Trypanosoma cruzi em diferentes hospedeiros do noroeste do Paraná, Sul do Brasil

Abstract

RESUMO: O objetivo deste estudo foi identificar as linhagens genéticas de Trypanosoma cruzi em 28 cepas isoladas de humanos (15), triatomíneos (9) e marsupiais (4) do Sul do Brasil. Os padrões de bandas das cepas foram comparados com as amostras referência: T. cruzi I (Sylvio e Co117G2), T. cruzi II (Esmeraldo e JG), T. cruzi híbrido (CL Brener, CAN III e SO3cl5) e T. cruzi III (222 e 231). Para realizar estas comparações foram analisados o polimorfismo na extremidade 3’ do gene ribosomal 24S? (rDNA), e o polimorfismo do tamanho do fragmento de restrição (RFLP) e seqüência da região 5’ subunidade 2 do gene mitocondrial da Citocromo Oxidase II (COII). O rDNA apresentou limitações para determinar as linhagens genéticas das cepas de T. cruzi, pois foram observadas bandas que estavam presentes em amostras referência de mais de uma linhagem. A análise do RFLP de COII revelou que para as cepas isoladas de humanos, uma pertencia à linhagem T. cruzi I, 13 pertenciam à T. cruzi II (IIb) e a análise da seqüência do gene de COII mostrou que uma cepa pertencia à linhagem T. cruzi III (IIc). As 4 cepas isoladas de reservatórios e cinco isoladas de triatomíneos foram definidas como T. cruzi I pela análise do RFLP de COII. Em quatro cepas isoladas de triatomíneos bandas das amostras referência T. cruzi I e II foram observadas concomitantemente, indicando infecção mista nestes hospedeiros. Os resultados mostram as limitações do rDNA e o potencial discriminatório de COII para indicar a presença de três linhagens genéticas de T. cruzi em humanos, T. cruzi I em reservatórios e triatomíneos e de infecções mistas em triatomíneos, indicando a importância do estudo para o entendimento da dinâmica de populações do parasito no Sul do Brasil

ABSTRACT: The aim of this study was to identify the Trypanosoma cruzi genetic lineages in 28 strains isolated from humans (15), triatomines (9) and opossums (4) in southern Brazil. The band patterns of these strains were compared with reference samples: T. cruzi I (Sylvio and Co117G2), T. cruzi II (Esmeraldo and JG), T. cruzi hybrid (CL Brener, SO3cl5 and CAN III) and T. cruzi III (222 and 231). To make these comparisons were analyzed the polymorphism in the 3’ end of 24S? ribosomal gene (rDNA), and the size polymorphism of restriction fragment (RFLP) and sequence of the region 5' of the 2 subunit gene of the mitochondrial Cytochrome Oxidase II (COII). The rDNA was limited to determine T. cruzi genetic lineages of strains, because was observed bands that were present in reference samples from more than a lineage. The RFLP analysis of the COII showed that to strains isolated from humans, one belongs to T. cruzi I lineage, 13 strains belong to T. cruzi II (IIb) and analyzing the sequence of the gene of COII, one strain has been shown to be belonging to the T. cruzi III (IIc). The 4 strains isolated from reservoirs and five isolated from triatomines were identified as T. cruzi I by analysis of RFLP of COII. In four strains of triatomines were observed bands of both patterns T. cruzi I and II, indicating mixed infection in this host. The results show the limitations of rDNA and the discriminatory potential of COII to indicate the presence of three genetic lineages of T. cruzi in humans, T. cruzi I in reservoirs and triatomines and mixed infections in triatomines, indicating the importance of study for understanding the dynamics of the parasite populations in southern Brazil
