dc.description.abstractRESUMO: A candidíase vulvovaginal (CVV) é uma infecção fúngica comum, afetando uma grande parcela das mulheres em idade reprodutiva. A CVV é causada principalmente pela espécie de levedura Candida albicans e, embora a infecção seja classicamente restrita à vulva e à vagina, é possível que os órgãos anexos sejam também atingidos pela levedura. Assim, este estudo teve como objetivo investigar a migração de C. albicans no trato reprodutivo feminino utilizando um modelo experimental murino. O estudo utilizou camundongos Balb/c em um modelo experimental previamente estabelecido para CVV. A infecção foi induzida por estrogenização com valerato de estradiol, seguida de inoculação vaginal com C. albicans. A progressão da infecção foi monitorada pela quantificação da carga fúngica e por análises histopatológicas dos órgãos reprodutivos internos (útero e ovários). Os animais foram eutanasiados em diferentes tempos pós-infecção para coleta de tecidos, que foram cultivados para quantificação fúngica e analisados histologicamente com colorações Grocott-Gomori e Hematoxilina-Eosina. No Artigo 1, foi demonstrado que a infecção vaginal por C. albicans resultou em elevada carga fúngica na mucosa vaginal desde o primeiro dia pós-infecção, com intensa descamação epitelial e infiltração inflamatória. A análise histopatológica revelou a presença de hifas fúngicas no epitélio vaginal e a disseminação de leveduras para o útero e os ovários a partir do terceiro dia de infecção, ainda que em menor intensidade do que no tecido vaginal. Essas observações confirmam a capacidade do fungo de migrar até o trato reprodutivo superior. No Artigo 2, o estudo aprofundou a investigação do envolvimento uterino, evidenciando alterações macroscópicas, como edema e hiperemia uterina, especialmente no terceiro e quinto dia pós-infecção. A histopatologia revelou inflamação progressiva, necrose epitelial e infiltração de neutrófilos, com redução da carga fúngica, após o sétimo dia. A presença de leveduras no tecido uterino sugere que a infecção vaginal pode induzir alterações uterinas e possivelmente afetar a fertilidade. Os resultados desta tese demonstram que C. albicans, quando inoculada no lúmen vaginal pode migrar para o útero e os ovários, provocando alterações histopatológicas significativas, bem como alterações no aspecto anatômico do útero. Esses achados inéditos ressaltam a importância de investigar as implicações clínicas da CVV além da infecção localizada, especialmente no contexto da saúde reprodutiva feminina, sugerindo a necessidade de mais investigações sobre suas implicações. Estudos futuros são necessários para avaliar a possível relação entre CVV e infertilidade, bem como os possíveis mecanismos imunopatológicos envolvidos.pt_BR
dc.description.abstractABSTRACT: Vulvovaginal candidiasis (VVC) is a common fungal infection that affects a large proportion of women of reproductive age. VVC is mainly caused by the yeast species Candida albicans and, although the infection is classically restricted to the vulva and vagina, it is possible that the accessory organs are also affected by the yeast. Thus, this study aimed to investigate the migration of C. albicans in the female reproductive tract using an experimental murine model. The study used Balb/c mice in a previously established experimental model for CVV. Infection was induced by estrogenization with estradiol valerate, followed by vaginal inoculation with C. albicans. The progression of infection was monitored through fungal burden quantification and histopathological analyses of internal reproductive organs (uterus and ovaries). Animals were euthanized at different post-infection time points for tissue collection, which were cultured for fungal quantification and analyzed histologically using Grocott-Gomori and Hematoxylin-Eosin staining. In Article 1, it was demonstrated that vaginal infection by C. albicans resulted in a high fungal load in the vaginal mucosa from the first day post-infection, with intense epithelial desquamation and inflammatory infiltration. Histopathological analysis revealed the presence of fungal hyphae in the vaginal epithelium and the dissemination of yeast to the uterus and ovaries from the third day of infection, although to a lesser extent than in the vaginal tissue. These observations confirm the ability of the fungus to migrate to the Upper reproductive tract. In Article 2, the study further investigated uterine involvement, evidencing macroscopic changes, such as edema and uterine hyperemia, especially on the third- and fifth-day post-infection. Histopathology revealed progressive inflammation, epithelial necrosis and neutrophil infiltration, with a reduction in the fungal load after the seventh day. The presence of yeast in the uterine tissue suggests that vaginal infection can induce uterine alterations and possibly affect fertility. The results of this thesis demonstrate that C. albicans, when inoculated into the vaginal lumen, can migrate to the uterus and ovaries, causing significant histopathological changes, as well as changes in the anatomical aspect of the uterus. These novel findings highlight the importance of investigating the clinical implications of VVC beyond localized infection, especially in the context of female reproductive health, suggesting the need for further investigation into its implications. Future studies are needed to evaluate the possible relationship between VVC and infertility, as well as the possible immunopathological mechanisms involved.pt_BR
